Well that really depends on a few factors:
1) Is she she a human giantess with no other extraordinary powers other then her size? The giantess is limited by what the amount their species can eat, so a naga giantess could swallow a same sized giantess just fine, but a human giantess couldn't unless there's something else going on.
2) The size of a the tiny being eaten. Your second post clears this factor up a lot sine you specify an amount, so no need to go into detail.
3) Has she trained herself to eat that much or not? I honestly don't know if it is possible to train yourself to handle that much food in one's stomach. And I really don't want to research too much into competitive eating and how much the world record holder managed to eat in one sitting and all that. But if it is possible, a trained giantess could eat more then an untrained one.
Depending on how the giantess ranks up with those factors, it could cause death.
Tl;dr: There are a few factors that come into play, but yes, it should probably kill a human giantess if they ate too many. That said the cancer/heart disease/whatever of eating us chemical filled tinies might kill them first. =p
